Retatrutide activates GIP, GLP-1 and GLUKAGON receptors, which are involved in controlling hunger and satiety, allowing people to feel fuller after eating for longer periods of time. This helps regulate blood sugar levels and leads to weight loss.
Eli Lilly announced Phase 2 test data for its new molecule called retatrutide, developed for the treatment of obesity. In the 24th week of the study, when retatrutide (1 mg, 4 mg, 8 mg or 12 mg) was administered to obese or overweight participants without diabetes, it was observed that the participants lost up to 17% of their weight on average. In the 48th week of the study, this rate increased to 24%.
Retatrutide's safety profile is similar to other incretin-based treatments. The most common side effects were gastrointestinal discomfort, which was usually mild to moderate and occurred during the dose escalation period.
Ania Jastreboff, Associate Professor of Medicine and Pediatrics at Yale University School of Medicine, noted that participants taking the highest dose of retatrutide lost 24.2% of their weight, and this rate continued to increase until the end of the study.
Retatrutide treatment was also associated with improvements in cardiometabolic measures such as systolic and diastolic blood pressure, triglycerides, LDL-cholesterol, total cholesterol, HbA1c, and fasting blood sugar and insulin.
Retatrutide works to treat obesity by targeting three different hunger-regulating hormones:
- GIP (gastric inhibitory peptide or glucose-dependent insulinotropic polypeptide) is an incretin hormone secreted from the intestines after a meal. It stimulates the beta cells in the pancreas to secrete insulin.
- GLP-1 (glucagon-like peptide-1) is an incretin hormone that stimulates the beta cells to secrete insulin.
- Glucagon is a hormone secreted from the alpha cells in the pancreas. It stimulates glucose production in the liver and helps maintain adequate plasma glucose concentrations.
Retatrutide has also shown positive results in the treatment of Type 2 diabetes, but is still in the research phase.
